Be careful when downloading from more cards than one at a time; you could easily get into trouble with file naming.

Even with Lexar's FW800 stackable, daisy-chainable readers I doubt you will win that much time since devices on the same IEEE1394 bus have to share bandwidth.

The same would be true if you were connecting two USB reader on two different USB bus (one in front of the machine and one on the back should work). But beware, if you have fast memory card (UDMA 4 or PIO6) and the reader that supports it, be sure to have a fast hard drive, because then everything will slow down again. Take the gold old watch and try to time everything if you get better result.

I agree with James. I use Backup and Burn. The software is great and handles 1 or more readers.
