The 5D Mk II will be a quantum improvement on the Nikon D300 simply because it is a later generation of technology, besides the larger format. The 5D Mk II with the 1.2 is arguably the best Canon has to offer in a normal lens, although you will get arguments about that.
However, since you are already invested in Nikon gear, you might consider taking a look at the Nikon offerings in full frame, the D700 and D3.
No matter what system you choose, there is always the risk that the results you got with the Hassy have a large subjective element in them, so you are basically faced with trying out the system you fancy. I would certainly recommend you make arrangements to try out the system of your choice first.
